Project opens and able to do everything without an issue - the problem is when trying to save we receive the following error message. Realize it is an issue with permissions on our system, but unable to determine what needs to be allowed so we can save? any ideas? Thanks!

Check please, if you have an acces to folder:
C:\Users\username\AppData\Local\Autodesk
To do'n lost data try

1) Save As -> save the file on your local drive
2) Run Fusion 360 with Admin righs (right-click on Fusion 360 Desktop Icon -> Run as administrator
3) Upload the file saved on the local drive as the new version and re-test "Save!
