Hello. I'm fairly new to AutoCAD, though I have been teaching myself how to use it here and there over the last year. I am more familiar with 3D CAD platforms like Fustion360 and SolidWorks.
Something that I cannot for the life of me is figure out how do apply a dimensional constraint to a construction line, so if for example, I want a rectangle's edge to be a set distance from a construction line, or if I am placing multiple construction lines in a layer and I want to have set spacing between them, when I try to use the Linear, Vertical, Horizontal, or Aligned dimensional constraints, it will not let me select a construction line, it tells me that it is an invalid selection. Is it not possible to apply dimensional constraints to/from construction lines?

No. Trim the construction lines so that they have a finite length and then you will be able to apply constraints.
