Just an FYI for you or others that might find this in the future, this thread was from 2006, and since that time Autodesk has added a built in options to disable the audio notification, which can be found by going to the Tools tab > Application Options button > Assembly tab > Relationship Audio Notification option

You've replied to a thirteen year-old posting. Inventor has had the option to turn off the constraint sound for many releases now (Application Options > Assembly tab > Relationship audio notification).

Hello, currently my coworkers computer keeps rechecking the sound affect, is there any reason/solution for this or should we look into creating a macro that automatically mutes inventor

The Application Options are saved in an xml file stored in %AppData% -> Autodesk -> Inventor 202x. It sounds that there is a system process resetting this user file regularly. Or, the user profile prohibits the user from saving such change.
Do a test. Try making the change and close Inventor. Go to %AppData% and check "UserApplicationOptions.xml" date. Is it recent?

it appears to only happen on randomly when constraining assembly (ex: click fastener first then assem reference= no noise ; but if we do this in reverse= noise)
