Please see Drawing 1 on A3.2 in the attached file. The annotations show in the cad file, they show in the print "thumbnail" but they do not show in the pdf. It's like they are set to defpoints, but they are not.

None of the xrefs are included...you may want to use the eTransmit command and create a zip file so it'll include all xrefs:

Your annotation layers are set to NOPLOT, perhaps you are not aware how that feature works (or perhaps exists): change that and your plotting issue will go away-
This is super amazing. I have never heard of it. In fact, my Layers window does not even show any such feature, and I am running AutoCAD LT 2025.
I attach a screen shot of my Layers window which looks different than yours. Why is that?

You are on a MAC (never mentioned it before now?), I'm not (and did not know about MAC), so this is the wrong forum for MAC based AutoCAD regardless of variants.
Your docked LAYER palette columns spacing is so wide you most likely just have it off-screen, isn't that what this button does? Or at least undock it.
Read all about NOPLOT in your program's HELP, here is a link to it online

for Autocad for Mac
is the Autocad for Mac board the better choice.
Perhaps the column is turned off ,
turn it on by right click on a column title like [Color]
