when creating a pdf plot, a filename is asked.
I want to automatically fill this filename.
Is this possible?
I am using acad 2006 and a .pc3 file.

Yes and No. What you do is set Acrobat to Automatically use the file drawing name.

Select printers from your start menu or Control Panel
Right Click on Adobe PDF then select properties
Select the Ports Tab
Click on Add Port and Select Any directory you want the files to go to. Check the box to the location you want.

--------
Now if you look at the general tab you will see Printing Preference button at the bottom of the page.
Click it and ...
Uncheck prompt for File Name
--------
Now you need to also set the Pc3 file to the same setting
In Autocad Goto File / plotter Manager
Double Click on the Adobe Pc3 file that you have previously created ... You did make one right?
then click on the last tab, Device and Document Setting, and click on Custom Properties and uncheck Prompt for File Name.

Now if will just create the PDF file in the directory with the current drawing file name and it won't prompt you.

It depends what PDF driver you are using but I would think all allow the
option to print to a file without prompting. We use PDF995 and that does but
it's configured extrenally, not by AutoCAD as it's just a generic printer
driver.
