I know how to do that.

1. Select the node. ("Selection > Select > Vertices")
2. Choose the type of load to apply. ("Setup > Loads")

Perhaps you are working on a CAD model and are really asking "how to create a node at a specific location so that I can apply a load". To do this, see the page "Construction Vertices - Seed Points" in the User's Guide.
