Space Separator works fine in floor plan, but option is grayed out in ceiling plan work set. Is there a way to use this option in the ceiling plan?

As you noticed this function is disabled in ceiling plans and therefore not available.

Place the space separator in a floor plan... the spaces can be recognized in a ceiling plan.

What doesn't work?  You can't draw Space separator in a floor plan?

I was trying to draw a space separator in a ceiling plan but it's not applicable yet in Revit! So, the only way is to use the Sketch ceiling?

Draw the lines in a floor plan, set the floor plan Underlay so that you can see the ceiling if you need it for reference to draw upon.
