Big assembly

How can i handel bigger assembly (more than 300 MB) in Inventor.
because everytime i received run time error in Inventor.

You can set the Level of Detail in the browser to suppress all components.
This may sound odd since nothing will be visible when you open an iam.
However, once the assembly is open you can unsuppress the part/subassemblies
you want to work on. By default all parts/subassemblies are loaded in memory
when you open an assembly.

I learned this at AU2006. I have some files that approach 600MB and applying
the above steps has helped me.

You may find that you can purge your IPT files for unused styles and
material. Starting templates with limited information will, one material,
with everything else purged will keep your file sizes smaller as well.
Having all the colors, styles, material only makes each IPT larger reducing
the IAM efficiency. It may not help that much now, but create a template for
each material you use, purge all unused data from the template. This will
reduce the file size of each IPT template. As you start building IAM with
1500-3000 IPT's it really start's to add up.
