.stl file disappears as soon as I open Simulation

Simulation does not support mesh geometry.  That is just an inherent limitation to the Simulation workspace currently

I’m following this video to simulate an object. 

Is there anyway I can simulate that stl file by converting it or something else?

I tried Mesh to Brep & after this the design went gray & now it can be opened in simulation.(though those random triangles are still there)

Should not be anything random.

All should be logical.

Can you File>Export your *.f3d file to your local drive and then Attach it here to a Reply?
