Hi, While using align option, when we try to align the second with the first object, the size of 2nd object gets modified. 

Is it not possible to align 2 objects without changing the object size?

Of course it is possible. What I'm seeing in your screen shot looks like you may be trying to align a rectangle. You need to remember that rectangle is four objects. You would need to move them in order to keep the rectangle the same size.

I want to align 2 rectangles, i.e., top line of 2 rectangles should be aligned in straight line. 

This type of alignment is possible & is shown in all videos, but I'm not getting. Why is it so?

It is not possible with the align command because rectangles are multiple objects. You can simply move the four lines to the perpendicular of the line in the other rectangle or you can make the rectangle into a single object in a number of ways depending on what you are trying to accomplish.

From your description it seems it has to do with the reference planes/lines that drive the rectangle family.

If the reference plane in the family is a Strong Reference, then aligning that object with that plane will stretch the family.

If the planes are Weak or No Reference, then the object will be moved.

It depends on the family you are trying to align and how it was built... instance parameters may mess with the size of your objects.
