Hi,
I am an architect and my team and I are using a shared Revit model to develop a design for a large and complex mixed-use building.
At the moment we are using AutoCAD to produced sketch designs before drawing the decided information in Revit. However, we would prefer to move out of AutoCAD completely. What is the best work process for carrying out sketch design options in Revit and subsequently updating the central model?

It depends on the project. But anyway, I strongly recommend learning all about you can do using Revit masses. Sometimes, you will not need it. But it is definitively one of the tools you should learn so that you can potentially use it during the concept design.

Depending on the project, you could also consider creating area plans to start off with. These allow you to shift the spaces around in 2D to get the basic outline. Then move into the 3d environment using massing if required to get the shape and feel of the building. You could also move straight into placing actual Revit elements (walls, floors, etc) if the building is simple enough.
Do you mean sketch options to explore alternative designs in the building? You can use Revit Design Options to actually model multiple options for different parts of the building all within the same project.
