Like many of us, I edit keynote files in Excel. Just upgraded to Revit 2017. 2017 won't reload a keynote file while it's open in Excel. This worked in all previous versions. Using Excel 2003.
To reproduce the error:
1) Open Revit
2) Open keynote file in excel.
3) Reload keynote file in Revit. It will reload fine.
4) Make a change to the keynote file in excel. Save the excel file (as .txt)
5) Reload keynote file in Revit. Get the error:
"Cannot open file D:\location\and\name\of\keynote\file.txt."
Works fine if the keynote file is open in notepad, which is lame.
You are correct in the behavior that you mention. R15 will open the file and reload it, R17 will give you the Warning Cannot open file (Excel 2016). something must have changed int he Keynote code that is causing the issue. Will have to check with the factory to see what the change might be.
Following up on this post. Please see the following information Keynote file won’t Reload in Revit 2017 with Excel open. I checked the internal case which is still ongoing, however, I suspect that a code fix will be required to address this. No date was given on when the changes will be made or released but if you keep and eye on the above link it will most likely be updated when this is resolved. I understand this is not ideal and I apologize for any inconvenience this is causing, but hopefully this will be addressed soon.

As alan.quinn mentioned it is a factory issue.
In the mean time here are few options for you
1. Use NotePAD++ , this is a awesome text editor. Also monitors if Network file has been changed and warns you to reload ever so often.
2. Libre Office? See if that helps.
3.Keynote manager . We have been using this program at our office for ages and this has been an awesome tool to manage your Keynote file.
at 49$ a seat , it is worth every penny. You can also get Concurrent license as well. Currently we are using his latest Cloud based Keynotes with C4R. Works great.. http://revolutiondesign.biz/keynotemanager/KMfeatures.html
Agreed it is not a solution , rather a work around.
Still I would say give Keynote manager a try, It is just awesome. (No I do not work for the company, but have been using Stephen's software for the longest time and it allows multiple people to work on the same file)
I have no problems editing the Keynotes txt file in Excel, saving it back to txt, CLOSING it from Excel, and loading it in (full) Revit 2017. Perhaps you have it open in Excel while loading it in Revit.
Arrgh, still no resolution. Seems to not be affected by Excel 2016.
