Revit 2021: what is the best practice to calculate the volume of a room?
In the screenshot below, I couldn’t figure out the best practice to calculate the volume of a room

Add a room and verify its base level and top height in the properties panel or in a section view. When you select the room, its volume should be visible in the Properties panel. Make sure that volume calculations are enabled in the Area Computations settings to display both areas and volumes.

I notice you are doing a Material Takeoff Schedule. Not clear what Room Volume has to do with that. Can you explain?
Also, you may look into Roombook Extension for Revit. It should be a part of your subscription benefits -- available through your autodesk.manage.com portal.

It works for me as a temporary solution. But what if you need to calculate the volume of the entire floor and then the entire building?
Then create a room schedule, because when you place more rooms you will be able to sort/group by level and calculate totals:
