We have just had a customer install this update and they can't open the application. It hangs on the splash screen?
try to install revit 2020.2 updates and see if helps. or do a clean uninstall and reinstall again. thanks
Did they follow the instructions given in the Release Notes? there are some files which need to downloaded and replaced manually … namely
DynamoRevitDS.sig and DynamoRevitVersionSelector.sig

I am also having this problem. I've tried on multiple machines, same thing.
I have pushed the Dynamo files on some test, but not in others. If you read the release notes, you only should have to do that if Dynamo doesn't show up after the 2020.2.1 update is complete. I pushed them just to be safe.
I'm not using Vault, so I didn't push that.
Thoughts?

To add to this, these are the methods I've attempted with no luck.
1. Push the .exe using PDQ with /quiet /norestart. Shows successful. Revit hangs on splash screen...
2. Extracted the contents of the .exe to .msi. There are (4)… Pushed again. Hangs...
3. Ran the .exe directly on a PC. Shows successful. Hangs...
4. Installed using Autodesk Desktop App. Hangs...
This is on all different machines. I'm running out of tricks!
I have ~ 200 installs running Revit 2020.2. There is no way on the planet I'm uninstalling and reinstalling.
When an Autodesk application hangs at the splash screen, there is a good chance it is a licensing issue that is holding things up. I would recommend checking the contents of the journals associated with these failed attempts to launch Revit. The information in the journal may point us in the right direction.
I would also ensure that the Autodesk Licensing Service is up and running on these systems. Also, if other Autodesk products are installed, can they be launched successfully? This will help to determine if the problem impacts all Autodesk products or just Revit.

We pushed that update yesterday and we didn't have any issues...do you have the latest version of Autodesk Licensing Service installed? might be it

I checked both LMTOOLS and adsklicensing - both are current.
LMTOOLS: v11.16.2.0
adsklicensing: v9.2.2.2501
I've attached a journal. It's pretty empty... Let me know if you can glean anything from that.
I also tried to open in Viewer Model. Same hang at splash screen.

Your journal indicates that Revit stopped loading just before the manage licensing call. The installation of the Revit 2020.2.1 Hotfix should not have made any changes that would have impacted licensing so the root cause of this problem is unclear.
My initial suggestion would be to reset the license method and see if that is enough to get Revit to load properly. Here are the steps to accomplish this task:
This action removes the currently saved license method (lm). The next time that Revit 2020 is launched, you should see the LGS dialog. If so, select the appropriate licensing method and see if Revit launches successfully at that point. If not, please collect and post a new journal file for review.

Unfortunately, it still isn't clear what is causing Revit 2020 to hang on these systems so we need to answer a few questions:
It would also be helpful if you could run the AdskLicensingInstHelper.exe tool again using the following syntax:
AdskLicensingInstHelper.exe list
This should echo back a list of installed Autodesk apps along with their current license configuration settings. Posting the results of this command may help our troubleshooting efforts.
Finally, I also recommend running the Revit repair process on one of the impacted systems and see if that makes any difference.

Network License on all the machines tested except 1 (mine). I tested on my laptop installing from the Autodesk Desktop App. I’m using Single User Sign-on and had the same problem.
Revit 2019 and AutoCAD are also using Network license.
Version prior to 2020.2.1 install is: 20.2.0.48
If I uninstall and reinstall 2020 and upgrade to 2020.2, it works again fine. I try to install the 2020.2.1 and it breaks again.

The output from the AdskLicensingInstHelper.exe list command indicates that licensing is properly configured for Revit 2020. If I read your previous post correctly, Revit is still functional after applying the 2020.2 Update but breaks after the installation of the 2020.2.1 Hotfix. If this is accurate, it sounds like your original deployment image is based on the original Revit 2020 full installer (or possibly the 2020.1 full installer). Along these lines, it would also be helpful to see a journal file from a Revit session prior to the installation of the 2020.2.1 Hotfix.
Another suggestion would be to eliminate the installation of the 2020.2 Update and apply the 2020.2.1 Hotfix directly to the version of Revit installed by the deployment. The 2020.2.1 Hotfix is a cumulative patch (it includes all the fixes and changes from previous updates and hotfixes) so there is no need to install the 2020.2 Update in this scenario.
It would also be helpful to get some additional details about your Revit 2020 deployment image. The first question is which media did you use to create this deployment image. Was it created from the Revit 2020 full installer or the full installer for a suite that includes Revit 2020 (Building Design Suite for example). Along the same lines, it would be helpful to know if there are any custom settings in this deployment image.

FYI - I just tried installing the new adsklicensing… 9.6.0.41. NO LUCK...
On PC #1, I did get prompted to select my licensing method. I chose Network and then it hung at the splash screen.
On PC #2, I did NOT get prompted and it just hung on the splash screen.
Install was done with Revit 2020 full installer. I've then over time updated to 2020.1 (when released), and then 2020.2 (when released) to stay current as Autodesk recommends. I'm now trying 2020.2.1...
Install came from AEC Collection.
EVERYONE has 2020.2... I tried uninstalling 2020.2 (which says it's successful), but 2020.2 still remains. I did it 2x using different uninstall methods with the same results. For grins, I tried the 2020.2.1 update even though Revit still reported 2020.2. I should have know better... Revit hung at splash screen.
How do I roll back 2020.2 to 2020.1 as you're suggesting? I guess I could try to go back to 2020 (with no point releases), but this is just muddying the water horribly. I should be able to push this update without having to uninstall these point releases on 300 computers.
I've now heard others are also having this same issue... so I'm glad it's not just me.
Can I suggest someone from Autodesk just give me a call instead of all this back and forth?
I do have an open ticket with Autodesk, but it's been unassigned now for 3 days. Ugh!
Ok, after all this back and forth, Autodesk support finally reached out to me. I was directed to attempt to open Revit 2020.2.1 on a PC that wasn't working so I could capture some log files, etc.. You won't believe this - Revit opens! I tested on a second computer and it opened too!
So, sorry to say, I have no clue why it's working now. Either Autodesk did some backend voodoo or our IT group did.
I did test the adsklicensing update (v9.6.0.41) earlier (which didn't work) as well, but my new PDQ push includes:
1. adsklicening (v9.6.0.41) update
2. Revit 2020.2.1
It's working.... go figure.
