I will work on a parcel subdivision with 20-30 lots of 4-5000 sqf each.
I need to define the street and lots layout, make some area calculations, mostly in 2D at this stage.
Why It doesnt let me create an area plan for the Site level?
Should I use Rooms or Area plans for the lots? What is a better way so to get some area schedules at the end?

Use property lines and a tag for them that shows the acreage of the enclosed area.

Regarding recommended workflow for parcel subdivision and improvements, we don't do this sort of planning directly in Revit and I don't think there is a "recommended workflow" for doing this sort of planning in Revit either. Generally, that planning is done by another software and discipline (e.g. Civil) and brought into the Revit Project environment.
Now, regarding your question: "Why It doesn't let me create an area plan for the Site level?", it's probably "Site" is a duplicate of another Level, such as Level 1. Look at the Properties of the "Site" Plan View and see what is listed for "Associated Level".
Lastly, regarding "Area or Room"; what specific information are you needing to report through this schedule? Knowing this will help me to help you decide.
As Barth said, you can create area plans, or if you want to dumb it down, you can create just filled regions and create tag family that tags their area.
I would like to tag Area, Mark (lot numbering) and maybe other custom parameters but mostly these two are most important.
You are right about the Site view, it is associated with Level 1.
