Our drawing index schedule has a parameter where "TBS" will blackout the unissued sheets.

We also have sheets that are not "TBS", and we also would like to black them out by conditional format.

I wonder if the "not equal to" condition is working, because when I change the condition to "not equal to"  "✓", as shown here, only the "TBS" be blacked out.

What happens if you put any value other than "✓" in the parameter for the ones currently showing blank?

Some parameters in Revit may look like they are blank but they are actually empty meaning nothing has been assigned at all and the conditional might not recognize that.

Enter something instead of Null value and see if they will be blacked out.
