I have download the soft copy(AutoCAD_2016_English_Win_64bit_dlm) from the product download.
Copy the files to usb flash drive.
During installation of AutoCAD 2016, it prompt "Please insert the disc AutoCAD 2016 Disc 1".
I am using usb flash drive for installation, the files downloaded from the product download is abt 4.88GB.
Please advised.

HMMM What kind of BS installer fail that it will not install from an external/removable storage device but that it will run from an internal drive.
How do those differ from a windows drive letter standpoint?
Each installation is unique to the users, machine, operating system, and settings. Sometime things happen and it just doesn't work properly. We are only users here and have no control over how the installer was created or why it works sometimes and sometimes it doesn't. All I can say if you are having issues with your installation to provide details and the installation log file.

I found, via Google, that adding "USE_REMOVABLE_DRIVE=YES" to the setup.ini (just under [SETUP]) solves it and lets you install from USB.
I had to do it twice as it seemed the setup reset the ini file. second time i marked the ini as read only and it worked fine.
I also had this problem like you. But i tried to extract it to a folder and it worked. Hope it's helpful for you.

Solution for 64 Bit Version
You can still take advantage of the setup.exe features and bypass the insert disk 1 issue by following these steps:
1- open the ISO file using any iso application.
2- extract the setup.ini file and open it in the notepad.
3- Using Notepad, edit the setup.ini file in the installer and add "USE_REMOVABLE_DRIVE=YES" under [SETUP].
Example:
[SETUP]
USE_REMOVABLE_DRIVE=YES
4- save the file and replace the setup.ini in the ISO image.
5- burn it or mount it to a virtual drive.
6- simply run setup.exe
this will make the setup.exe works fine and continue the process.
It works for Naviswork Freedom installation as well.
Message: Please insert disc: NW
