I am trying to make a vacuum wand and there is a feature that requires a loft. I understand there is a centerline loft and a rails loft. Selecting one or the other has an impact on the shape generated by the loft tool. It is essential that I use the centerline method, however Fusion 360 tells me that I do not have a centerline that intersects all the profiles I am trying to loft. I have 3 profiles that I sketched and two fully constrained lines. One goes from profile 1 to profile 2 and the other goes from Profile 2 to Profile 3. How can I turn these two lines that are fully constrained into a single line so that I can loft with the centerline? Or is there a simpler way around this?

Your centre line segments must be in same sketch, be coincident connected, and smooth, (tangent connections), for it to work.
Selecting the Rail is done by accumulating all the segments.
Checking close on the photo, seems like your not selecting the centre line, should be blue, however there is a blue circle in profile three being labelled as centre line?...
Otherwise I would need to check the file, to see what else may be in play.

Can you File>Export your *.f3d file to your local drive and then Attach it here to a Reply for demonstration using your actual geometry.

A centerline does not need to be a single line. It looks, from this image, as if you have not selected what I expect you want as a centerline:
