basic cap and thread question

Prototyping a medical device on autodesk 123d. Printing threaded port of entry into machine for which I also made a cap. I used the loft function to build threading. Do I need to scale up the cap (female) to allow for tolerance? The cap is the negative impression of the threaded barb.
