Any idea what would cause this to suddenly happen to my layer property manager? Everything is workable via other methods, but it is a bit of a pain. Already re-booted.

Try to use LAYERDLGMODE set to 0.
It'll be old school, but it might work. I actually run my layer window this way. AutoCAD seems to be more responsive this way, but slower when dealing with the actual layers so there's a trade off.

Save your profile/workspace. Changing to another one and then back should fix it but it won't be permanent.

Well, the third re-boot did it. Not sure why, we may never now. I will keep the suggestions for other type issues.
