If by " align the UCS to a three-dimensional SPLINE" you mean define a UCS that has it origin at the start of the spline and it z axis tangent to the spline at its beginning then (without the use of LISP) you can use the ZA option of the UCS command.  Input the first Control Vertex for the origin and the second CV for the second point.  You can use LIST to get the locations of the CVs.
