this can have a lot of reasons..

 

One of reason we can check this way:

I think your files are on a network location?

Restart AutoCAD.

Use Open command to open the main file, look at the time.

Hiw much?

start etransmit and setup an etransmit package as zip,

Settings:

No bind option.

All xref files to the same folder.

Close ACAD

Unzip this ZIP in a local folder,

all files incl. xrefs are there.

start the main file from this folder.

Same time as the original file?

 

If yes.

Close ACAD, Export your custom setting (you know how?)

Reset ACAD to default

Start ACAD again open the local mainfile.

Same time as before?

Then Import your exported settings back.

What is the productversion? (command About)

About RAM, check the taskmanager or resourcemanager to see what happen while open the file,

if ACAD need 50% of Ram and 50%~ is free, it isn't a ram problem.
