You can purge all unused Layers at once if you answer No to the question "Verify each name..."
You can also use wildcards when AutoCAD prompts "Enter name(s) to purge <*>:"
For example you can purge all unused layers witch names begins with number 1 - you need to use 1* wildcard for this.
