I would like to be able to highlight different areas in a drawing. Just like using a highlighter pen. Anyone know how to do this?

in short, I've created a "highlight" layer, give a tolerable transparency, then just draw a polyline with a thickness

I tried this and only generate a thin yellow polyline around selected object. I would like this closed polyline filled with yellow? I attempted to use the hatch feature, but could get it to work.

The result I was trying to get you to do was more like a thick polyline with a global width that would be sufficient to cover the object, having a color as desired, together with a transparency of like 50 or so...

PLINE objects can be assigned a global width to be as wide as you need: give it a try.
You can also use SOLID command to create a filled rectangle: you can also just draw a rectangle and fill it with a solid batch for the same effect.
