Yes.

Civil 3D labels are designed to be used in the way you describe. You create label styles, assigning text heights for your various label components. Those label automatically display to the (specified text heights x viewport scale). As you change viewport zoom scale, your labels will automatically re-size for the current scale. You can have multiple viewports zoomed to multiple scales. All labels will be the same "plotted" size.

Problem is once you regenerate (rea) your layout all the labels get back to the size of model space!!

That depends on your zoom scale within the viewport. If you want to be able to zoom in without them changing size, for when you're designing and labeling, lock the viewport scale (this is done from Paperspace). Even when you go into modelspace, the zooming will happen in paperspace, so the relationship between your labels and the viewport will not change.

I think @Anonymous wants the same label to display a different size in different viewports. For instance in viewport 1 the label must be 2.5mm high in paper space and in viewport 2 it must be 3.5mm high in paperspace.

If that is the case, then unfortunately, Civil 3D does not work that way. The label will always scale to the viewports scale. You will need to create seperate label styles with different text heights for each height you want displayed and put them on different layerrs and then freeze the layers in the relavant viewport.
