You can change the "reference station", but reversing the direction is
currently not supported. The reference station is defined on the Alignment
Properities dialog.

I tried that. The new alignments in Civil 3d do not seem to follow the direction of the polyline. They appear to go left to right with north being up. If you "Create by Layout..." you can force the direction of the alignment.
