It is possible to define your own subassembly from a polyline, if that's
what you meant. If you meant that you want to combine several "BasicLane"
subassemblies into one subassembly with editable pavement depths, then you
will need to do this with VBA. Check to see if there is an existing
subassembly that looks like you want it to. Some of the more advanced
Roadway subassemblies (such as the superelevation subassemblies) allow you
to specify multiple pavement depths. With the more advanced templates, if
you don't provide certain optional parameters, the default behavior is
sufficient in many cases.
If you want to create a simple subassembly using a polyline, the size and
shape are fixed and constant (ie: if you want to modify one of the
dimensions, you have to redraw the polyline). Check out Corridors->Create
subassembly from polyline. The process is a little tricky at first, but
it's easy once you get the hang of it.

I can create a subassembly for each layer of my pavement structure but
how I combine them so they are one subassembly like the subassemblies
shipped with Civil 3D.
