I have made a Curve label style as shown in the attachment. I have to add this as a multiple segment label, which does not update when I extend or shorten my alignment. I want to use the label style in a label style set for an alignment. So how do I do that?

It is not possible to add detailed geometry information in the label sets in they what you show in the image.
So that kind of sums it up.
I am a bit confused, I can label my alignment and have those labels update if I happen to alter the alignment. How are you labeling the alignment?

Not sure what this is about. You can add the curve rad as shown quite easily in civil 3d.
To add a label style to an Alignment Label Set you need to create the label style in the Alignment - Label Styles area.
You can make the curve radius label style under Alignments -> Label Styles -> Curve
To get the label to work on lines, you will need to make a false radius label style under Alignments -> Label Styles -> Lines
Just set the display component to R = (infinity symbol)
Then just add your alignment curve label style and alignment line label style to a new alignment label style set.

I'm labeling my alignment using the "add labels" -> "Multiple segment" from the contextual ribbon of the alignment. Yes they update via the alignment at minor changes, but have you tried adding geometry or deleting a curve and adding one again?
If you have a workaround that is easier than; "delete labels" and add multiple segments again I would like to know about it.
