I have this same problem with profiles all the time. I create a new design profile and the toolbar is completely grayed out. There is no way to create any geometry for the profile, so I have an empty profile that cannot be edited. I have to close the drawing and reopen it, delete the empty profile, and then recreate it in order to fix it.
I have seen this when there are multiple drawings open and the Alignment Creation Tools toolbar is open for another alignment, in one of the other drawing files.
