1) Operating system?
2) AutoCAD "flavor" (if applicable)?
3) What level of service pack are you running?
4) Processor?
5) Graphics card and driver version?  Is this driver version the Autodesk certified version?  http://usa.autodesk.com/adsk/servlet/syscert?siteID=123112&id=18844534
6) System memory?
7) HDD or SSD?  What is your autosave time set to?
😎 Are you using blocks?
9) Are you using xrefs?
10) Are you turning off layer as you work so only the layers you currently need are active?
11) What shademode are you working in?
12) Have you already purged, audited, and otherwise cleaned up the drawing?
13) Are there levels of detail in your drawing that can be simplified that you don't really need?

AutoCAD is generally limited to 1-core or thread of your processor, except for 2D regen and rendering.  So you want that 1 core to be really fast (3GHz+ base clock minimum).  Of course, a good graphics card is also key when working with these large models.
