I'm having this same problem although I might have a cause for me atleast.

Running 2010
I haven't got Raster design installed and never have.

I tracked it down to this sequence.

I copied an alignment from one drawing into another. (same drawing just slight different road layout)
Created the profile
All acting fine.

I then edited the profile by deleting the PVI's. (still lets me save)
I then add 2 new points then it won't let me save

I tried to delete the alignment but it crashed the software. I think my situation is different from the rest but I thought I would add it to the mix

It's happen to me before and at that point you have no other way of saving the file you've lost what ever you have done before the last save. Now what I do is to save the file like every 5mins and now if you run into that problem again you don't lose that much work. It seems like a lot of work but in time you don't even know your saving the file.

The other thing that it might be is do you have xref files in this drawing if so detach, purge, audit and then save. If that is the case you have to go to each file that was xrefed and do a purge and audit then save.

I have been using AutoCad 2009 for the past year or so and all of a sudden started getting this error today on multiple drawing files, some of which haven't been used in several months. I tried everything I read above and nothing seemed to work consistently. What is most interesting is that if I move the files off the server and work on them locally I don't have any saving problems. It only seems to give me the error when working off of dwg files off of the server...

I can't make any sense of it but am curious if this "work around" might help the other folks who are struggling with the same issue.

I can generate this error just by switching between layout tabs and trying to save each time.
-open drawing
-save works fine
-click on a layout tab
-save works
-click on another layout tab
-save starts to generate the error and will not save again until the drawing is re-opened but appears to be saving even though it spits out an error.

??? ya got me???

had the error on one machine, user had the autosave directory set to read-only. Make sure all directories that require writing (autosave, menus, pgp, temp, logs, etc.) have that ability.

I've had the autosave directory set read-only for a couple of years. I've never had this problem when Raster Design wasn't loaded. I think there may be multiple causes for this symptom.

hmmm... if mine is set to read-only, it won't save and I get the same error noted here.

I verified the read-only. One of the wonderful vagaries of AutoCAD!

so your AutoCAD writes to a directory that is marked read-only??
