Hi,

Is there anyway to design a RC beam with holes? E.g 300x600mm beam with 150mm diameter hole.

Yes, this can be done provided the beam is created in the RC Beam stand alone module. For a beam imported from a model it is possible to adjust the distribution of stirrups.
