Is there a way to change the format of alignment chainage from 1501+120 to this format 1501km120?

I don't see a way to change the abbreviation for the plus sign. But in your label, you should be able to add a Station Value with the Output set to "left of station character". Then type "km" after that field. Then add a Station Value with the Output set to "right of station character". This will form the three parts of your label.

Troma beat me to it but here is the caption we use for Imperial and we have ours split out as he describes and use a block for the "station character" = km in your case.
