I reset the defaults, and I actually had to copy and paste a number of files from the AutoCAD CD onto my C drive.
I can no longer remove/repair/reinstall the software. It won't let me do any of those actions.
When I try to start the program, the error message is: "Customzation File load failed. File not found: acadlt." I click OK and the program closes.
It sounds like you need a clean reinstall at this point. It might be best to post a new thread over on the installation forums, but here are the instructions:

My IT department ended up re-imaging my computer. After that point, I was able to install AutoCAD and use it without a problem.

AutoCAD 2012 hangs on startup for me, on an intermittent basis. One of the regular fixes for me is to go to task manager - services, and turn off the print spooler till it starts. Another one is to clear the temp folder. Obviously I'd rather fix the problem for good, but it does the trick for me.
